It refers to the first card among all the [Specified Slot] affinity cards that you use from your hand this turn after this [When Played] ability activates. Even if you use a card without the [Specified Slot] affinity before using the first one with it, that first [Specified Slot] affinity card's AP cost is still reduced by 1.
